儀器描述

Fluidigm C1™ Single-Cell Auto Prep System是一台全自動單細胞分離與全基因庫製備系統，藉由拋棄式微流道晶片針對單一細胞進行獨立分離，於晶片中可完成單細胞裂解、核酸純化、反轉錄作用和擴增放大流程，最後將單細胞之終產物流放至獨立槽中，進行後續分析。

主要特點

  • 1. 透過微流道系統，每次可以分離96 或 800顆細胞
  • 2. 全自動細胞內容物萃取，有效降低細胞處理所需的時間
  • 3. 微流道室體積小，可以有效降低試劑所需費用
  • 4. 搭配客製化程式與試劑可以進行染色體，核酸，蛋白質分離
  • 5. 可以同時分析染色體序列與基因表現分析，ATAC-seq 與基因表現分析
  • 應用範圍：單一細胞基因表現(Single-Cell Gene Expression)、mRNA定序、DNA定序、表觀遺傳學或miRNA表達檢測。
  • 樣品型式：均質懸浮細胞
  • 晶片選擇: 針對不同細胞大小有3種尺寸 (耗材自備)

Description

Fluidigm C1 is an automatic single cell isolation and whole gene library auto preparation system. The integrated fluidic circuits (IFC) can automatic capture single cell and perform cell lysis, RNA isolation, reverse transcription, and pre-amplification. The final product can be export to individual well for library preparation or other application.

Main specification

  • 1. The IFC can isolate 96 or 800 cells per chip
  • 2. Automatic cell extraction which decrease cell handling time
  • 3. Small volume which large decrease enzyme and reagent cost
  • 4. Isolation of genomic DNA and RNA in the same cell
  • 5. Perform ATAC-seq and gene expression in the same cell
  • Application: Single-Cell whole transcriptome analysis、gene targeting analysis, genomic DNA seq, miRNA seq, ATAC-seq.
